Cinnamaldehyde Decreases the Pathogenesis of Aeromonas hydrophila by Inhibiting Quorum Sensing and Biofilm Formation

چکیده

Antibiotics were the main fishery drugs for treating Aeromonas hydrophila (A. hydrophila) infection, which would generate selective pressure and result in appearance of antibiotic-resistant bacteria. The bacterial quorum sensing (QS) system provides a new alternative strategy against A. infection. QS inhibitors can reduce virulence behaviors by disrupting QS, has no effect on growth. Therefore, we studied cinnamaldehyde from natural plant extract aiming to its pathogenicity. efficacy was evaluated various aspects, including effects aerolysin, lipase, protease, swarming motility, biofilm formation, acyl-homoserine lactones (AHLs), QS-related genes. Moreover, therapeutic vitro vivo studied. results showed that could decrease phenotypes regulated QS. transcriptions related genes (aerA, ahyR, ahyI) downregulated following addition cinnamaldehyde. assays show aerolysin-mediated A549 cell injury increase survival rate crucian carp infected with hydrophila. These indicate be candidate inhibitor

Subtilosin Prevents Biofilm Formation by Inhibiting Bacterial Quorum Sensing.

Subtilosin, the cyclic lantibiotic protein produced by Bacillus subtilis KATMIRA1933, targets the surface receptor and electrostatically binds to the bacterial cell membrane. In this study, subtilosin was purified using ammonium sulfate ((NH4)2SO4) precipitation and purified via column chromatography. Subtilosin's antibacterial minimum and sub-minimum inhibitory concentrations (MIC and sub-MIC)...

Trigonella foenum-graceum (Seed) Extract Interferes with Quorum Sensing Regulated Traits and Biofilm Formation in the Strains of Pseudomonas aeruginosa and Aeromonas hydrophila

Trigonella foenum-graecum L. (Fenugreek) is an important plant of the Leguminosae family known to have medicinal properties. However, fraction based antiquorum sensing and antibiofilm activities have not been reported from this plant. In the present study T. foenum-graecum seed extract was sequentially fractionated and sub-MICs were tested for above activities. The methanol fraction of the extr...

Quorum sensing-dependent regulation and blockade of exoprotease production in Aeromonas hydrophila.

In Aeromonas hydrophila, the ahyI gene encodes a protein responsible for the synthesis of the quorum sensing signal N-butanoyl-L-homoserine lactone (C4-HSL). Inactivation of the ahyI gene on the A. hydrophila chromosome abolishes C4-HSL production. The exoprotease activity of A. hydrophila consists of both serine protease and metalloprotease activities; in the ahyI-negative strain, both are sub...

Influence of polyphenols on bacterial biofilm formation and quorum-sensing.

Many bacteria utilize sophisticated regulatory systems to ensure that some functions are only expressed when a particular population density has been reached. The term 'quorum-sensing' has been coined to describe this form of density-dependent gene regulation which relies on the production and perception of small signal molecules by bacterial cells. As in many pathogenic bacteria the production...

Small molecule inhibitors of bacterial quorum sensing and biofilm formation.

Bacteria monitor their local population densities using small molecules (or autoinducers) in a process known as quorum sensing. Here, we report a new and efficient synthetic route to naturally occurring bacterial autoinducers [N-acyl l-homoserine lactones (AHLs)] that is readily amenable to the synthesis of analogues. This route has been applied in the first synthesis of a library of non-native...
